      DescriptionCaspase 2, recombinant human active
OverviewCaspase-2 (also known as ICH-1, NEDD-2) is a member of the interleukin-1beta converting enzyme (ICE) family of cystein proteases. Similar as other caspases, caspase-2 also exists in cells as an inactive proenzyme. During apoptosis, procaspase-2 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases. The processed form of caspase-2 consists of large (19 kDa) and small (12 kDa) subunits which associate to form the active enzyme. The active recombinant human caspase-2 was expressed in E. coli. The expressed caspase-2 spontaneously undergoes autoprocessing to yield the subunits characteristic of the native enzyme. The recombinant caspase-2 preferentially cleaves the substrates consisting of consensus sequence VDVAD (e.g., VDVAD-AFC and VDVAD-pNA). The recombinant enzyme can also be used as a positive control in caspase assays or in determining the specificity of caspase substrates.Recommended usage: 0.5-1 unit for fluorometric caspase assays and 2-4 units for colorimetric caspase assays. Optimal working dilutions must be determined by end user.
      Product Information
PresentationPartially purified recombinant caspase 2 with a full length HIS-6-tag. Approximately 10-20% caspase 2 protein. The remainder is E.Coli proteins. Approximately 0.8-2 mg total protein/U of activity; E.Coli proteins have no reactivity. Lyophilized. Reconstitute in 25 μL PBS containing 15% glycerol.

      Biological Information
PurityPartially purified recombinant caspase 3 with a full length HIS-6-tag. Approximately 10-20% caspase 2 protein. The remainder is E.Coli proteins. Approximately 0.8-2 mg total protein/U of activity; E.Coli proteins have no reactivity.
Specific Activity25U: Where one unit of the recombinant caspase-2 is the enzyme activity that cleaves 1 nmol of the caspase substrate VDVAD-pNA (pNA: pnitroanaline) per hour at 37oC in a reaction solution containing 50 mM Hepes, pH 7.2, 50 mM NaCl, 0.1% Chaps, 10 mM EDTA, 5% Glycerol, and 10 mM DTT.
      Entrez Gene Number
Entrez Gene SummaryThis gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. The proteolytic cleavage of this protein is induced by a variety of apoptotic stimuli. Alternative splicing of this gene results in multiple transcript variants that encode different isoforms.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P42575 # Involved in the activation cascade of caspases responsible for apoptosis execution. Might function by either activating some proteins required for cell death or inactivating proteins necessary for cell survival.
SIZE: 452 amino acids; 50685 Da
SUBUNIT: Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a p18 subunit and a p12 subunit. Interacts with LRDD.
TISSUE SPECIFICITY: Expressed in larger amounts in the embryonic lung, liver and kidney than in the heart and brain. In the adults higher level expression is seen in the placenta, lung, kidney, pancreas than in the heart, brain, liver and skeletal muscle.
PTM: The mature protease can process its own propeptide, but not that of other caspases.
SIMILARITY: SwissProt: P42575 ## Belongs to the peptidase C14 family. & Contains 1 CARD domain.
