DescriptionMacrophage-Colony Stimulating Factor Protein, Recombinant human
OverviewM-CSF is a potent hematopoietic factor produced by a variety of cells including lymphocytes, monocytes, fibroblasts, endothelial cells, myoblasts and osteoblasts. It is involved in development, differentiation, and survival of blood monocytes, tissue macrophages and their progenitor cells. The human is reactive in murine systems, but the murine molecule exhibits no activity on human cells.

Human M-CSF is a 36.8 kDa disulfide-linked homodimer composed of two 159 amino acid polypeptide subunits.

      Applications
ApplicationThe M-CSF protein is a potent hematopoietic factor produced by a variety of cells including lymphocytes, monocytes, fibroblasts, endothelial cells, myoblasts & osteoblasts.
      Key Applications
      • Cell Culture
Application NotesFor most in vitro applications, M-CSF exerts its biological activity in the concentration range of 0.1 to 20.0 ng/mL.

      SourceE. coli
      Specific ActivityThe ED50 as determined by a cell proliferation assay using murine M-NFS-60 cells is < 5 ng/mL, corresponding to a specific activity of > 2 x 105 units/mg.
      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a cytokine that controls the production, differentiation, and function of macrophages. The active form of the protein is found extracellularly as a disulfide-linked homodimer, and is thought to be produced by proteolytic cleavage of membrane-bound precursors. The encoded protein may be involved in development of the placenta. Four transcript variants encoding three different isoforms have been found for this gene.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P09603 # Granulocyte/macrophage colony-stimulating factors are cytokines that act in hematopoiesis by controlling the production, differentiation, and function of 2 related white cell populations of the blood, the granulocytes and the monocytes-macrophages. CSF- 1 induces cells of the monocyte/macrophage lineage. It plays a role in immunological defenses, bone metabolism, lipoproteins clearance, fertility and pregnancy.
SIZE: 554 amino acids; 60119 Da
      SUBUNIT: Homodimer or heterodimer; disulfide-linked.
SUBCELLULAR LOCATION: Cell membrane; Single-pass membrane protein (By similarity). & Processed macrophage colony-stimulating factor 1: Secreted, extracellular space (By similarity).
PTM: Glycosylation and proteolytic cleavage yield different soluble forms. A high molecular weight soluble form is a proteoglycan containing chondroitin sulfate. & Isoform 1 is N- and O-glycosylated. Isoform 3 is N- glycosylated.

      Storage and Shipping Information
Storage ConditionsThe lyophilized protein is stable for a few weeks at room temperature, but best stored at -20° C. After a quick spin, reconstitute in water to a concentration of 0.1 - 0.5 mg/mL. Reconstituted M-CSF should be stored in undiluted aliquots at -20° C. Avoid repeated freeze/thaw cycles. This solution can then be further diluted into other buffers and stored at 2° - 8° C for one week or stored at -20° C for future use.
