Not Western Blot
Not Neutralization Studies
Application NotesFlow Cytometry (see comments)
Immunoblotting (not recommended)
Immunofluorescence (see comments)
Immunoprecipitation (10 µl/mg protein lysate)
Neutralization Studies (not recommended)
Application CommentsDoes not cross-react with c-erbB1 (EGFR), c-erbB2/HER2/neu or c-erbB4/HER4. It does not block the binding of heregulin to c-erbB3, has no effect on tyrosine kinase activity, receptor internalization or tumorigenic growth of cells overexpressing c-erbB3. This antibody is particularly well suited for immunoprecipitating the unoccupied as well as the occupied receptor. This antibody has also been reported to work for flow cytometry and immunofluorescence. Antibody should be titrated for optimal results in individual systems.

DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with specified immunogen and fusing splenocytes with NSO mouse myeloma cells. Recognizes the ~185 kDa c-ErbB3 protein.
BackgroundErbB3/HER3, a member of the epidermal growth factor receptor (EGF-R) family, was originally isolated as a cDNA clone from a human carcinoma cell line. The molecule is transcribed from a 6-kilobase mRNA, which can be identified in a variety of different tissues. Epidermal growth factor (EGF) initiates its effects on cell growth through interaction with a 170,000 dalton cell surface glycoprotein receptor. Binding of EGF, amphiregulin, or transforming growth factor-alpha (TGF-α) to the receptor or to other members of the receptor family activates a tyrosine-specific protein kinase intrinsic to the EGF-receptor protein family. While there is a large degree of conserved homology between the cytoplasmic regions of EGF-R and HER2/neu, the HER3 molecule is more unique. This observation suggests that HER3 is subject to regulatory signals distinct from those affecting EGF-R and HER2/neu and, therefore, signals an alternate biochemical response. In particular, it has been shown that the ligand for HER3 is NDF/heregulin a property shared with HER4. c-ErbB3/HER3 is overexpressed in a variety of tumors (e.g., breast, stomach, pancreas and colon).
