Background InformationMultiple epidermal growth factor-like domains protein 10 (UniProt; Q6DIB5; also known as Multiple EGF-like domains protein 10, Megf10) is encoded by the Megf10 gene (Gene ID: 70417) in murine species. Megf10 is a type I transmembrane membrane receptor protein that is involved in phagocytosis by macrophages and astrocytes of apoptotic cells. Megf10 is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 26-857), a transmembrane protein (aa 858-878), and a cytoplasmic domain (aa 879-1147). It also contains 15 EGF-like domains in the extracellular domain. Its expression is observed in developing myoblasts, muscle stellate cells, stellate cells of the cerebellum, kidney, and macrophages. It is expressed at the cell surface in clusters around cell corpses during engulfment. It serves as a receptor for C1q that binds phosphatidylserine expressed on the surface of apoptotic cells. It is shown to be essential for astrocyte-dependent apoptotic neuron clearance in the developing cerebellum. Megf10 also plays a role in muscle cell proliferation, adhesion, and motility and is shown to be an essential factor in the regulation of myogenesis. It controls the balance between skeletal muscle satellite cells proliferation and differentiation through regulation of the notch signaling pathway. Mutations in Megf10 gene are reported to cause early onset myopathy, areflexia, respiratory distress, and dysphagia. (Ref.: Saha, M., et al. (2107). Human Mol. Gen. 15(1); 2984-3000).
