Panel Overview
Conditions that affect the heart or vasculature are termed cardiovascular diseases (CVD) and include hypertension, congenital heart disease, and coronary heart disease. As a group, CVDs are a heterogeneous set of conditions with numerous causative and accessory factors.
Atherosclerosis (narrowing and hardening of the arteries), internal bleeding and blood clots, inflammation, infection, genetic and environmental factors underlie many CVDs and are the subject of large and growing research and therapeutic efforts. Atheromas, or plaques, are composed of a matrix of immune cells, fatty deposits, and extracellular matrix proteins. Atheromas can restrict blood flow, causing angina, and also may become unstable and break loose, which can block arteries resulting in heart attack or stroke. The initial stages of atheroma formation involve the attachment of leukocytes to the endothelial surface. Selectins are transmembrane carbohydrate-binding proteins that mediate the initial weak binding of immune cells to the endothelium. The attachment is strengthened by subsequent interactions involving intracellular adhesion molecules 1, 2 and 3 (ICAM-1, 2, 3) and vascular cell adhesion molecule 1 (VCAM-1). Osteopontin expression by smooth muscle cells near the atheroma promotes retention of macrophages. Similarly, leptin induces expression of monocyte chemotactic protein 1 (MCP-1). Ligand binding to the receptor for advanced glycation end-products (RAGE) contributes to the inflammatory response through the expression of MCP-1 and VCAM-1.
The BeadPlex<® Human CVD Panel 3 is designed to detect any or more of the following analytes associated with human cardiovascular disease in suspension:
• Soluble E-selectin (sEs)
• Soluble Intercellular Adhesion Molecule-1 (sICAM-1)
• Leptin
• Osteopontin
• Soluble P-selectin (sP-selectin)
• Soluble Receptor for Advanced Glycation Endproducts (sRAGE)
Panel Components
Storage: All components of the BeadPlex Human CVD Panel 3 are shipped at 2 to 8°C. Upon receipt, store all kit components at 2 to 8°C. Do not freeze.
The BeadPlex Human CVD Panel 3 is available in two kit configurations:
• Complete Kit
• Mix and Match Format.
For utmost convenience, end-users may order the BeadPlex Human CVD Panel 3 - Complete Kit (Cat. No. BPHCVD03-6). This kit contains all respective capture beads and all components in the BeadPlex Human CVD Panel 3 - Base Kit. Desired capture beads will need to be pre-mixed prior to running the assay. The kit contains enough components to assay one 96-well plate.
For maximum flexibility, end-users may configure their own customized multiplex assay through the Mix and Match Format. This format consists of a BeadPlex Human CVD Panel 3 - Base Kit (Cat. No. BPHCVD03-BASE) and a customized choice of any or all analyte-specific capture beads as listed in Technical Bulletin (TB535). The Base Kit and selected components contain enough reagents and buffers to assay one 96-well plate.
