      DescriptionAnti-triphospho-PER2 Antibody (Ser662/Ser665/Ser668)
      Alternate Names
      • Period circadian protein homolog 2
      • hPER2
      • Circadian clock protein PERIOD 2
Background InformationPeriod circadian protein homolog 2 (PER2), or Circadian clock protein PERIOD2, is a component of the circadian clock mechanism which is essential for generating circadian rhythms. It is a negative element in the circadian transcriptional loop by Influencing clock function by interacting with other circadian regulatory proteins and transporting them to the nucleus. PER2 negatively regulates CLOCK NPAS2-BMAL1 BMAL2-induced transactivation. This gene is a member of the Period family of genes and is expressed in a circadian pattern in the suprachiasmatic nucleus, the primary circadian pacemaker in the mammalian brain. Genes in this family encode components of the circadian rhythms of locomotor activity, metabolism, and behavior.

      Control
      • Untreated and lambda phosphatase treated HEK293 cell lysates transfected with Myc-tagged triphospho PER2 (Ser662/Ser665/Ser668).
PresentationPurified rabbit polyclonal in buffer containing PBS with 0.05% sodium azide and 50% glycerol.
      Applications
ApplicationDetect PER 2 using this rabbit polyclonal antibody, Anti-triphospho-PER2 Antibody (Ser662/Ser665/Ser668) validated for use in western blotting.
      Key Applications
      • Western Blotting
Application NotesWestern Blotting Analysis: A representative lot detected triphospho PER2 (Ser662/Ser665/Ser668) in Flag-tagged triphospho PER2 (Ser662/Ser665/Ser668) transfected HEK293T cell lysates (Hutchinson, J. A., et al. (2011). J Biol Chem. 286(10):8688-8696.).
      Biological Information
ImmunogenLinear peptide corresponding to human triphospho PER2 (Ser662/Ser665/Ser668).
      HostRabbit
SpecificityThis antibody demonstrates cross reactivity against phospho RPS6 (Ser247) (Hutchinson, J. A., et al. (2011). J Biol Chem. 286(10):8688-8696.). This antibody requires that all three amino acid sites (Ser662/Ser665/Ser668) be phosphorylated for the detection of phospho-PER2 (Ser662/Ser665/Ser6668) (Shanware, N. P., et al. (2011). J Biol Chem. 286(14):12766-12774.).

Quality AssuranceEvaluated by Western Blotting in untreated and lambda phosphatase treated HEK293 cell lysates transfected with Myc-tagged triphospho PER2 (Ser662/Ser665/Ser668).

Western Blotting Analysis: A 1:250 dilution from a representative lot detected Myc-tagged triphospho PER2 (Ser662/Ser665/Ser668) in 10 µg of untreated HEK293 cell lysates, and demonstrated a loss of signal in 10 µg of lambda phosphatase treated HEK293 cell lysates.

      Storage and Shipping Information
Storage ConditionsStable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
