      Background InformationHeterogeneous nuclear ribonucleoprotein U (UniProt Q8VEK3; also known as hnRNP U, Nuclear matrix protein sp120, SAF-A, Scaffold attachment factor A) is encoded by the Hnrnpu (also known as Hnrpu, SAFA, Sp120) gene (Gene ID 51810) in murine species. The scaffold attachment factor A (SAF-A) is a component of heterogeneous ribonucleoprotein (hnRNP) particles. About 50% of the protein is tightly associated with nuclear matrix. The N-terminal region of hnRNP U/SAF-A interacts with A/T-rich areas of double-stranded DNA that is also known as the scaffold attachment regions. SAF-A is also known to use its C-terminal RGG domain for targeting G/U-rich heterogeneous RNA. The ability of hnRNP U/SAF-A to bind to both DNA and RNA implicates a critical role of SAF-A in diverse nuclear functions, including transcriptional regulation, telomere length control, and X chromosome inactivation (XCI).
