      Background InformationThe protein named Transient receptor potential cation channel subfamily V member 3 or TrpV3, or alternatively Vanilloid receptor-like 3 (VRL-3) and encoded by the human gene TRPV3 is a non-selective cation channel important in temperature sensation and vasoregulation. TrpV3 is expressed for instance in the sensory neurons of the skin and is activated by temperature. TrpV3 also plays a role in keratinocytes proliferation in hair growth; TrpV3 suppression promotes growth, over activation results in apoptosis and hair follicle regression. Indeed TrpV3 is often deemed the most important TRP (transient receptor potential) channel in keratinocytes of the skin. TrpV3 is widely expressed in the CNS and in the keratinocytes of the skin and epithelial tissues. In the brain TrpV3 channels appear to be important for mood regulation and TrpV3 appears to modulate some anti-inflammatory responses in the brain mediated by the anti-inflammatory agent incensole acetate. TrpV3 is also regulated by calcium, ATP and calmodulin, and while it is considered a temperature sensor, recent research challenges that somewhat but TrpV3 still plays important roles in other skin functions such as cutaneous sensations, hair development and barrier function, and mutations in TrpV3 have been linked to a rare genodermatosis known as the Olmsted syndrome.
