      Background InformationTumorous imaginal disc 1 (Tid1) is a regulatory protein, comprised of a CR-type zinc finger and a J domain, belonging to DnaJ family of cochaperones/specificity factors for the Hsp70 family. Tid1 is involved in regulating several cellular processes such as cell proliferation, induction of apoptosis, and cell survival. Although mainly localized to mitochondria, most of the functions performed by Tid1 are nonmitochondrial. Tid1 induces the release of mitochondrial cytochrome C and activates caspase 3. In humans, Tid1 expression produces two splice variant isoforms that each serve a contrasting role in apoptotic regulation; Tid1-L (43 kDa) upregulates apoptosis in response to either mytomyocin C or TNF, and Tid1-S (40 kDa) serves to suppress apoptotic activity. The highest levels of expression have been observed in the liver, lung, heart and skeletal muscle, as well as in keratinocytes. Originally, it was described as interacting with the HPV E7 oncoprotein.
