      Background InformationPyruvate kinase PKLR (EC; also known as Pyruvate kinase 1, Pyruvate kinase type L, Pyruvate kinase isozyme R/L, Pyruvate kinase isozymes L/R, Pyruvate kinase isozymes R/L, R-type/L-type pyruvate kinase, Red cell/liver pyruvate kinase, Pyruvate kinase, liver and blood cell) is encoded by the PKLR (PK1, PKL, PKR, RPK, PKRL) gene in human. Pyruvate kinases are cellular glycolytic enzymes that mediate glucose metabolism. There are four mammalian isoforms encoded by two gnens. The L gene is alternatively spliced to produce the L (liver) and R (erythrocyte) isoforms, while the M (muscle) gene is alternatively spliced to produce the M1 or M2 isoform. Pyruvate kinase M1 (PKM1) is predominantly expressed in adult tissues, while PKM2 is expressed during embryonic development. Tumor cells express exclusively PKM2. Defects due to PKLR gene mutations are the common cause of chronic hereditary nonspherocytic hemolytic anemia (CNSHA or HNSHA).
