      Background InformationPARP3, or Poly ADP ribose polymerase 3, also known as ADP-ribosyltransferase diphtheria toxin-like-3 (ARTD3), IRT1, NAD(+) ADP-ribosyltransferase 3 (ADPRT-3), Poly-ADP-ribose-synthase 3 (pADPRT-3) and encoded by the human gene named PARP3/ADPRT3/ADPRTL3 is a key enzyme involved in DNA repair and surveillance. PARP3 catalyzes the poly ADP-ribosylation of a limited number of acceptor targets that are involved in chromatin architecture and DNA metabolism; the modification by PARP3 is required for proper recognition of DNA damage and appears to be an enzymatic from DNA surveillance and mitotic fidelity checkpoint. PARP3 interacts with several proteins including PARP1, PRKDC, and XRCC5 and PARP3 is a component of the centrosome as well. PARP3 is localized primarily in the nucleus except during times of cell division. PRAP3 is expressed in all tissues with the highest levels being in the kidney, skeletal muscle and heart. Moreover different family members play different DNA repair roles in the cell. Recent research demonstrates that PARP3 is active in the efficient joining of DNA double-strand breaks during non-homologous end-joining whereas PARP1 modulates UV mediated DNA damage, and PARP3 has been identified as a critical player in the stabilization of the mitotic spindle and in telomere integrity notably by associating and regulating the mitotic components NuMA and tankyrase 1 and thus is a target for cancer therapy as well. Interestingly too, PARP3 is also implicated in ADP ribosylation of key signaling molecules in other cellular systems such as energy metabolism, neural crest development and memory. EMD-Millipore’s Anti-PARP3 rabbit polyclonal antibody has been tested in western blot on human kidney tissue lysates and in paraffin embedded immunohistochemistry on human pancreatic cancer tissue sections and in fluorescent immunocytochemistry on HeLa cells in culture.
