      Background InformationMucins are high molecular weight glycoproteins that are heavily glycosylated with oligosaccharide side chains linked O-glycosidically to the protein backbone. At least nine mucin genes have been identified to date. The pattern of tissue and cell-specific expression of these mucin genes suggests a distinct role for each member of the diverse mucin family. In epithelial cancers, many of the phenotypic markers for pre-malignant and malignant cells have been found on the carbohydrate and peptide moieties of mucin glycoproteins. Deregulation of tissue and cell-specific expression of mucin genes has also been described in epithelial cancers. At present, the role of mucin glyoproteins in various states of epithelial cell carcinogenesis, including the pre-neoplastic state and metastasis, is under investigation.

      Application NotesWestern blot:Clone CLH2 shows a weak reactivity with a high molecular weight smear characteristic of mucin from a native mucin extract from normal gastric mucosa and from gastric diffuse carcinoma. Typical dilution is 1:100.

      SpecificityAntibody MAB2011 is specific for the MUC5AC tandem repeat of human mucin. No cross reactivity with other peptides from MUC2, 3, 6, or 7.

      Immunoreactivity of MAB2011 has been detected in the following normal human epithelial tissues: bronchus, endocervix, stomach, and gallbladder. The predominant staining pattern in normal tissues is perinuclear. Antibody reactivity has been observed in foveolar and mucopeptic neck cells of normal gastric mucosa. The expression of MUC5AC has been frequently observed in gastric carcinomas and may be used as a marker of gastric differentiation.

      The CLH2 antibody shows a weak reactivity with a high molecular weight smear characteristic of mucin from native mucin extract from normal gastric mucosa and from gastric diffuse carcinoma .

      The antibody was generated by immunizing mice with a synthetic tandem repeat peptide The antibody recognizes the sequence TTSTTSAP within the tandem repeat of MUC5AC (1). It further recognizes glycosylated as well as unglycosylated MUC5AC.

Highly expressed in duodenal Brunner's glands, gall bladder, seminal vesicle, pancreatic centroacinar cells and ducts, and periductal glands of the common bile duct.
      DEVELOPMENTAL STAGE: Early expressed in fetal development and was observed in Brunner's glands and pancreatic ducts at 18-19 weeks and in gastric glands at 20 weeks of gestation. Expressed transiently in the nephrogenic zone of the kidney in the early mid-trimester of development. Detected in the epithelium of ureteric buds at 13 weeks and at lower levels from 17 to 23 weeks of gestation.
