      Background InformationMitochondrial peptide methionine sulfoxide reductase, also known as, Peptide-methionine or S-S-oxide reductase or Peptide Met(O reductase or Protein-methionine-S-oxide reductase or PMSR, and encoded by the gene name(s), MSRA, catalyzes the reduction of methionine sulfoxide to methionine. It is a widely expressed enzyme that constitutes an important repair mechanism for oxidatively damaged proteins that accumulate during aging and in certain degenerative diseases. hMSRA displays high expression levels in the detoxifying organs kidney and liver, but an equally high expression is also found in cerebellum and hippocampus. Although considered as a mitochondrial enzyme, a cytosolic form has also been reported in rat tissues. Rat MSRA contains two initiation sites of translation, corresponding to Met-1 and Met-2. It is suggested that cytosolic form results from translation from Met-21, whereas the mitochondrial enzyme is synthesized as a precursor protein starting at the Met-1 initiation site and is then processed during import into the mitochondria. Silencing of MSRA gene results in loss of mitochondrial membrane potential and increased reactive oxygen species production in human lens.
