      Background InformationThe enzyme currently named Iron-sulfur cluster assembly enzyme ISCU, mitochondrial, or in earlier literature NifU-like N-terminal domain-containing protein or NifU-like protein, and encoded by the human gene named ISCU or NIFUN, is a biosynthetic enzyme involved in the assembly of iron-sulfur proteins. There are two isoforms in mammals. Isoform 1 is localizes to the mitochondria, whereas isoform 2 localizes to the cytoplasm. ISCU is expressed in a variety of tissues including brain, heart, skeletal muscle, lung and kidney. Iron-sulfur proteins are proteins characterized by the presence of iron-sulfur clusters containing sulfide-linked centers in variable oxidation states. Iron-sulfur cluster proteins are best known for their role in the oxidation-reduction reactions of mitochondrial electron transport. Both Complex I and Complex II of oxidative phosphorylation have multiple Fe-S clusters and thus require functional ISCU to exist. In addition, iron-sulfur clusters are found in a variety of metalloproteins, such as the ferredoxins, as well as NADH dehydrogenase, hydrogenases, Coenzyme Q - Cytochrome C reductase, Succinate-coenzyme Q reductase and nitrogenase. New research has demonstrated that iron-sulfur proteins also can regulate gene expression and can be regulated by nitric oxide. EMD-Millipore’s Anti-ISCU rabbit polyclonal antibody has been tested in western blots on mouse heart tissue extracts, paraffin embedded immunohistochemistry on human lung cancer sections, and in fluorescent immunocytochemistry on MCF7 cells in culture.
