      Background InformationHelicase POLQ-like (EC; UniProt Q8TDG4; also known as Mus308-like helicase, POLQ-like helicase) is encoded by the HELQ (also known as HEL308) gene (Gene ID 113510) in human. HELQ is a DNA helicase that translocates in the 3 to 5 direction and can displace oligonucleotides of 70 nt or more from DNA. HELQ opens up the parental strands at a blocked DNA replication fork and remodels nascent lagging strand intermediates to facilitate the loading of subsequent factors required for processing DNA damage or restarting DNA replication. HELQ is associated with the RAD51 paralogs RAD51B/C/D, XRCC2, ATR, and operates in DNA repair and cell cycle checkpoint signalling in response to DNA interstrand crosslinks (ICLs). HELQ gene distruction in human cells enhances cellular sensitivity and chromosome radial formation by the ICL-inducing agent mitomycin C (MMC). HELQ-knockout cells show reduced phosphorylation of the ATR substrate CHK1 as well as a reduced accumulation of G2/M cells in response to MMC treatment. Helq-/- mice exhibit subfertility, germ cell attrition, ICL sensitivity and tumour predisposition.
