      Background InformationFBXO7, also known as F-box only protein 7, and encoded by the gene name FBXO7/ FBX7, is a substrate recognition component of a SCF (SKP1-CUL1-F-box protein) E3 ubiquitin-protein ligase complex that mediates the ubiquitination and subsequent proteasomal degradation of target proteins. FBXO7 recognizes BIRC2 and DLGAP5. FBXO7 has been shown to play a role in the clearance of damaged mitochondria via selective autophagy (mitophagy) by targeting PAEK2 to dysfunctional depolarized mitochondria. FBXO7 promotes MFN1 ubiquitination. FBXO7 interacts via its C-terminal proline-rich region with DLGAP5. FBXO7 interacts with CDK6, BIRC2, and promotes its interaction with D-type cyclin. FBXO7 isoform 1 interacts with PARK2 via the n-terminal ubl domain. FBXO7 isoforms that lack the ubiquitin-like domain do not interact with PARK2. FBXO7 isoform 1 and isoform 2 interact with PINK1 via N-terminal region; however isoform 3 does not interact with PINK1. In addition, FBXO7 interacts with PSMF1. Mutations in FBXO7 have been associated with Parkinson disease 15 (PARK15), a neurodegenerative disorder characterized by parkinsonian and pyramidal signs, including clinical manifestations, such as: tremor, bradykinesia, rigidity, postural instability, spasticity in the lower limbs, and hyperreflexia.
