Background InformationCD151 is a tetraspanin and is thought to aid in the creation of multi-protein complexes at the surface of cells. CD151 forms secure laminin-binding compounds in the kidney, using integrins alpha3beta1 and alpha6beta1, and in the skin, using alpha3beta1 and alpha6beta4. CD151 may also play an important role in the inner ear and may also be involved in erythropoiesis. CD151 may be an important marker for prostate cancer and the higher the level of CD151, the less likely the patient survives. Research has shown that mice lacking CD151 have a damaged re-epithelialization process and lack wound healing ability.

      • Immunofluorescence
      • Immunohistochemistry
Application NotesImmunofluorescence Analysis: A previous lot was used by an independent laboratory in IF to show colocalization of CD151 with laminin receptors a3b1 and dystroglycan in the glomerulus and with a6b1, dystroglycan and the Lutheran molecule in tubular epithelial cells (Sterk, L., et al. (2001). J. Cell Sci. 115:1161-1173).

Immunohistochemistry Analysis: A previous lot was used by an independent laboratory to stain CD151 in kidney, skin and heart tissues (Sterk, L., et al. (2001). J. Cell Sci. 115:1161-1173).

Immunoprecipitation Analysis: A previous lot was used by an independent laboratory in IP (Sterk, L., et al. (2001). J. Cell Sci. 115:1161-1173).

      EpitopeAlpha 3 Binding Site
SpecificityThis antibody recognizes CD151 alpha 3 binding site.
The antibody does not recognize CD151 when bound in complex to integrin alpha3beta1 or alpha6beta1 (Sterk, L., et al. (2001). J. Cell Sci. 115:1161-1173).

      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein that is known to complex with integrins and other transmembrane 4 superfamily proteins. It is involved in cellular processes including cell adhesion and may regulate integrin trafficking and/or function. This protein enhances cell motility, invasion and metastasis of cancer cells. Multiple alternatively spliced transcript variants that encode the same protein have been described for this gene. [provided by RefSeq].

      UniProt Number
UniProt SummaryFUNCTION: Essential for the proper assembly of the glomerular and tubular basement membranes in kidney.

      SUBUNIT STRUCTURE: Interacts with integrins alpha3beta1, alpha5beta1, alpha3beta1 and alpha6beta4, with CD9 and CD181.

SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.

TISSUE SPECIFICTY: Expressed in a variety of tissues including vascular endothelium and epidermis. Expressed on erythroid cells, with a higher level of expression in erythroid precursors than on mature erythrocytes.

POLYMORPHISM: CD151 defines the MER2=RAPH1 antigen of the RAPH blood group system. 92% of Caucasians are MER2-positive and 8% are apparently MER2-negative.

IINVOLVEMENT IN DISEASE: Defects in CD151 are the cause of nephropathy with pretibial epidermolysis bullosa and deafness (NPEBD) [MIM:609057]. NPEBD is characterized by the association of hereditary nephritis, epidermolysis bullosa, deafness, and beta-thalassemia minor.

SEQUENCE SIMILARITIES: Belongs to the tetraspanin (TM4SF) family.
