      Background InformationBarrier-to-autointegration factor (BANF1, BAF), also known as Breakpoint cluster region protein 1 (BCRG1), plays a critical role chromatin organization as well as subsequent gene expression particularly in embryonic stem cells (ESCs) development where it is required to maintain self-renewal potential. BANF1 initial naming comes from its interaction with viral proteins during infection. Various retroviruses, including HIV-1, make use of BANF1 protein to protect their DNA from normal autointegration destruction functions, and thus allowing viral integration in the host DNA chromosome. BANF1 is localized primarily in the nucleus but can be found in the cytoplasm as well, but during mitosis it is specifically localized to the chromosomes. The protein can bind dsDNA and nuclear membrane proteins and is thought to also play a role in chromatin positioning with respect to the nuclear periphery. There appear to be multiple isoforms as well. EMD-Millipore’s Anti-BANF1 rabbit monoclonal antibody has been tested in western blot on HeLa, HepG2 and 293T human cell lysates and in immunohistochemistry on paraffin embedded thyroid gland carcinoma tissue.
