The Stericup vacuum filtration system combines a Steritop filter unit with a receiver flask for processing and storing volumes from 150 mL to 1,000 mL.

The Stericup vacuum filtration system combines a Steritop filter unit with a receiver flask for processing and storing volumes from 150 mL to 1,000 mL.

Its new no tip/easy grip flask design and compact profile improve stability during filtration, make gripping the receiver easier, and make Stericup Filter Units ideal for use in laminar flow hoods.

As an added convenience, the bottom of the receiver flask is slightly recessed, enabling capped flasks to be stacked for convenient storage.

To determine their effects on Mouse Stem Cell growth or differentiation, three lots of Stericup-GP devices were used to filter media with LIF. Once filtered, this media was used to passage mouse stem cells five times to verify that Stericup-GP filtration does not impact pluripotency of mouse stem cells.

    QuestionAnswer
    Are Stericup and Steritop units autoclavable?No. The highest temperature that the product can be used at is 50C ( 122F ).
    Can I bubble point my Stericup/top?No. The unit is not designed to handle the pressures required to perform a bubble point test. It is designed to be run under vacuum only (15 psi max). Bubble points for any of the membranes in the Stericups/tops need to be run at pressures ranging from 22 psi to 57psi.
    Can I filter 100% serum with the Stericup?Yes. The Express membrane is the highest throughput membrane available on the market today.
    Can I purchase extra receiver flasks for the Stericups separately?Yes, the catalog number is SC00B10RE.
    Can you freeze Stericup units?You can successfully freeze and store many aqueous solutions ( such as culture media ) at temperatures to -20C ( -4F ). We strongly recommend that you run a trial under actual conditions to test for suitability of the bottles for frozen storage.
    Do you have a filter for 100% serum?Yes, you can filter serum with Millipore Express membrane. The Express membrane was designed for this application and offers fast filtration time. Millipore Express is available in a large number of filter devices including Millex syringe filters, Stericup and Steritop vacuum devices, Sterivac for large volumes of media and Steriflip for 50 mls.
    For tissue culture media filtration which membrane should I use?The best membrane for this application is the Millipore Express membrane. It was designed specifically to filter tissue culture media with serum. It is low protein binding, has fast flow and high throughput (which is also called filter capacity).
    I want to sterilize 100% ethanol. Are any of the Stericups or Steritops compatible with ethanol?The funnel and receiver flask is made of polystyrene. Any molded polystyrene will craze in pure ethanol and could structurally fail depending on the forces being applied to it. The funnel could handle the short exposure time but the receiver would craze eventually. Therefore we would not recommend the Stericup with 100% ethanol. The Steritop, if you keep the exposure time to the funnel minimal, should be fine. As you dilute the alcohol the compatibility improves.

    Both Express and Durapore membranes are compatible. MCE is not.
    I will be filtering tissue culture media with your Stericup/top and am concerned that wetting agents in the filter could effect my cells. Which membrane type would you recommend to avoid this?All of Millipore's Tissue Culture Media filtration sterilization devices contain membranes that are wetting agent (surfactant ) free.
    Is the Express membrane surfactant free?Yes, the Express membrane is surfactant free. It also has a very low extractables specification - less then 1.5% by weight.
    When I use my Stericup/top, sometimes some of my sterilized solution gets pulled into my vacuum line. How can I eliminate this from happening?Attach your vacuum tubing and turn the vacuum on before adding the liquid to the Stericup/top. This eliminates any initial solution that flowed through the membrane before starting the vacuum from being pulled into the vacuum line.
    What is the typical holdup volume for Stericup.The hold up volume is about 6 ml. If you give the unit a slight shake, you can reduce the holdup volume by about 60% to 2.4 ml. Solutions containing protein have a slightly higher holdup volume, about 7 ml, again reduced to less than half by a slight shake.
    What is the diameter of the membrane in the Stericup and Steritop units?All of the cups and tops currently have a 70mm diameter membrane, which has 43 cm2 of surface area.
    Why use a vacuum driven filter rather than a pressure driven filter?Unlike positive pressure filtration, vacuum driven filtration requires only a vacuum source. It does not require pressure vessels or peristaltic tubing pumps. Stericup and Steritop units are pre-sterilized and disposable making setup and cleanup easy!
    Should I filter my acrylamide solution prior to gel casting?Yes, filtering your acrylamide solution before adding the polymerization activator ammonium persulfate and TEMED will accomplish two things.
    1. It will eliminate particulate matter that may interfere with base calling when sequencing or that may create "hot" spots in the gel, and
    2. It will aid in degassing, which will promote efficient polymerization of the gel.

    Products appropriate for this application would be Stericup, Steritop or Steriflip with express membrane.
    It sounds like air is leaking into my Stericup, what should I do?First make sure that the top is securely fastened to the base. Discontinue use if there appears to be any cracks on the apparatus.
    What Stericup would you recommend to remove mycoplasma?The Stericup VP ontains an 0.1um Express membrane. The VP membrane has been validated for the removal of mycoplasma.
    Which pre-filter should I be using with my Stericup or Steritop unit?You should use an AP20 75mm pre-filter (AP2007500). To help keep the filter from floating in the solution to be filtered, insert the pre-filter under the pre-filter tab inside the Stericup or Steritop.
    What is the expiration date on Stericup and Steritop units?The expiration date, which specifies product sterility, for the Stericup and Steritop products is three years from the date of manufacture.
    What type of flow rates can I expect with the Stericups and Steritops?For typical non -serum tissue culture media you can expect flow rates from 1-1.5 lpm.
    What diameter prefilter is used in the Stericup?We recommend a 75mm diameter. Even though the membrane has a 70mm diameter, there is a very thin lip of plastic that seals it in place in the funnel. The 75mm gives it a nice snug fit.

    Inside the cup there is also a small tab of plastic just above the filter designed for you to slip the prefilter under. This eliminates the prefilter floating up when you add your solution.
    What is the thread size on the Stericup units?The thread size on the Stericup units is 45mm.
    Are Stericup/tops compatible with formamide?Yes. Formamide is probably the least organic compound. It's almost like water. So it would be compatible with everything. The potential mistake is mistaking formamide for dimethylformamide, which is a powerful organic solvent.
    Can I sterile filter my 10% acetone, 90% water solution through a Stericup with the Durapore membrane?The membrane will be OK since it is well supported in these configurations ( Durapore is not compatible with 100% acetone). The molded plastic components (polystyrene/polyethylene) may have a tendency to develop microcracks, or crazing, due to the acetone being able to releave the stresses from the molding process. If it does craze this will reduce the strength of the molded parts. Crazing would be instantaneous with high levels of acetone. At 10% it is difficult to say. If under a strong vacuum the crazed part could even fail.
