|Can I use compressed air instead of nitrogen to pressurize the cell?||We don't recommended it because air may cause pH shifts (C02 adsorption) or oxidation of product.|
|Can the membranes for the Stirred Cells be reused?||Yes, the membranes can be reused several times given they are used, cleaned, and stored properly. The operating instructions for the Ultrafiltration Membranes (literature number 99101) includes a restoration protocol for the membranes specific to each type. Also, under the operating procedures section is the proper method for usage and storage.|
|Can the Stirred Cell products be sanitized with Sodium Hypochlorite?||Yes. We recommend 100 ppm (parts per million) of Sodium Hypochlorite for 30 min.|
|Does tubing come with the stirred cell?||Yes, the following tubing comes with the stirred cell: a. Four feet of 1/4" pressure tubing for pressurizing the stirred cell. b. Four inches of permeate tubing.|
|How do I know the membrane in my stirred cells is in correctly?||The glossier side of the membrane should be facing up.|
|How do I remove the stirred cell after I've vented the pressure?||Push the cap down, then slide the cell out from the retaining stand.|

|I bought some replacement bodies for my old style stirred cell. They did not fit properly. Has there been a product change?||In March of 1990 the stirred cell bases were changed to a white plastic molded part from a black metal part. Bodies compatible with the black metal base are not compatible with the white plastic base. When ordering a replacement body you will also need to order a new base. Here are the catalogue numbers for the affected products:
|In what order do I assemble the parts to the tube fitting assembly?||Assemble in the following order: First, the hexagonal nut slides on to the red pressure tubing. The metal grab ring with flange facing away from hex nut fits into hex nut. The white plastic spacer with countersink surface towards grab ring is next. Finally, the o-ring is placed onto the end of the assembly.|
|What comes with the 8000 series stirred cells?||Everything except the magnetic stirrer, pressure source and the membrane.|

|To what protein concentration can the solution be concentrated to using a stirred cell?||Practical limit is 10% by weight at room temp and 55psi. Slightly higher if solution can be warmed.|

|What concentration can I use with the Stirred Cells?||All standard stirred cells operate within the following range:
|What could be wrong if the stirrer assembly in the Stirred Cell won't spin?||The stir bar won't spin unless the stirred cell is pressurized.|
|Are the Stirred Cells autoclavable?||Yes, stirred cells can be autoclaved at 121 C for 30 minutes. It is important, however, to tighten the base the loosen 1/4 turn before autoclaving. The tubing and the tubing connector are not autoclavable.|
|Are there cases where a stirred cell outperforms a crossflow type system?||Yes, at protein levels below 10 ug/ml. Also, pressure has a greater effect on flux as protein concentrations decrease. Most laboratory tangential flow systems operate at low pressure (20-25 psi), due to peristaltic pump limitations. Stirred cells do not require a pump for operation. They are normally pressurized to 55 psi.|
|Do you sell parts for the Stirred Cells individually? What is the part number and price?||Yes, the parts lists for the Model 8000 Stirred Cells can be found in the Stirred Ultrafiltration User's Guide P99228. The parts list for the Model 2000 High Output Stirred Cell can be found in the user's guide P99109. Parts for the older style Stirred Cells (Model 12, 52, 202 or 402) are no longer being replaced.|
|I don't have any flow in my stirred cell. What happened?||a. Cell is not pressurized.
b. Membrane upside down. Glossy side should face up.
c. Plastic sheet not removed from membrane (XM-300 only),
d. Solution may be too concentrated (viscous).
e. Check that the membrane holder and filtrate port is not blocked.
|Where can I order the filtrate tubing to the Stirred Cell (Cat. No. 4500430) when it is no longer offered at Millipore?||Cat. #4500430, TUBING (TYGON R-3603, 5/32X3/32), can be purchased through Fisher Scientific (800-766-7000) or Cole Palmer (800-323-4340)|
|Should I use the Ultracel regenerated cellulose membrane or the Biomax membrane in my TFF system?||It depends on the nature of your process volume and the process speed required. The Ultracel membrane is a lower protein binding membrane that should be used in low protein loading applications(<20 g/m2). The Biomax membrane is a faster flowing membrane that should be used in high protein loading applications (>20 g/m2). Ultracel is less tolerant to extreme pH conditions than the Biomax, but the Ultracel is more compatible with organic solvents.|
|What products and catalog numbers are required for the diafiltration set-up?||The diafiltration set-up requires the following:
1. Stirred Cell, for example catalogue number 5121 - Model 8010 (which uses 25mm membrane discs).
2. Magnetic stirring table.
3. Reservoir or dispensing pressure vessel. For example, catalogue number 6028 - RC800 Reservoir (800mL) or catalogue number XX6700P01 - 3.785 liter Dispensing Pressure Vessel (this will require relief valve, hose connectors, etc.)
4. CDS10 Concentration/Dialysis Selector, catalogue number 6003.
5. Pressure tubing to connect the system to a pressure source. XXPE00010 is 10 feet of 1/4" OD polyethylene tubing.

|What fitting do I need to connect the stirred cell to a regulator?||Tubing adapter kit (product number 6022) provides two fittings that should fit any regulator.|
|What pressures are Stirred Cells operated at?||The maximum operating pressures for Models 8003, 8010, 8050, 8200 & 8400 is 75psi and Model 2000 is 100 psi. The recommended operating pressure for all Stirred Cells is 55 psi and <10psi when using a 100,000 MWCO membrane or greater.|
|Can the stirred cell be autoclaved with a membrane in place?||Yes, but only with the YM membrane. Put a small amount of buffer in the device to keep the membrane wet, tighten the bottom then loosen 1/4 turn. Remove any tubing and fittings, they are not autoclavable.|
