      Catalogue Number AB5922
      Brand Family Chemicon®
      Trade Name
      • Chemicon
      Description Anti-Nestin Antibody
      Alternate Names
      • Nestin
Background Information Nestin, a large intermediate filament protein (class Type VI) expressed during development and in myotendinous and neuromuscular junctions. Nestin expression is restricted, typically disappearing by E18. Nestin is thought to be a reasonable neuronal marker; however, recent studies have found nestin expression in other cell types such as endothelial cells {Folia Biologica, 44(5):155, 1998}. Nestin identifies the most primitive neuroepithelium but also identifies many other embryonic tissues, so it is not specific for CNS. Nestin expression is seen in almost all GBMs (Glioblastoma mulitformes) and many melanomas (both primary and metastatic) but not in any metastatic carcinoma.

      Control
      • Positive controls are U251 or U373 human glioma cell lines. Detects a ~200-220 kDa protein, note slightly difference sizes for nestin are quoted in various paper and from various cell lines
      • in U251 (Human Glioma line) nestin migrates at ~260 kDa.
      • in WI-38 (human lung line) reduced nestin migrates at ~220 kDa
Presentation Rabbit polyclonal Serum in buffer containing Liquid.
      Applications
Application Anti-Nestin Antibody is an antibody against Nestin for use in IC, IH, IH(P) & WB.
      Key Applications
      • Immunocytochemistry
      • Western Blotting
      • Immunohistochemistry (Paraffin)
      • Immunohistochemistry
      Application Notes Western Blot:
1:1,000-1:5,000 dilution of a previous lot was used on cultured cells or human brain tissue. It is suggested that the membrane be blocked with PBS containing 5% nonfat milk and 0.05% Tween. The antibody may also be diluted in the same blocking buffer.

      Immunocytochemistry:
1:200 dilution of a previous lot was used on cells fixed in 4% paraformaldehyde and permeabilized with 0.02% Triton.

      Immunohistochemistry(paraffin):
1:200 dilution of a previous lot was used on formalin fixed, paraffin embedded tissue sections.
      Biological Information
      Immunogen Fusion protein.
Concentration Please refer to the Certificate of Analysis for the lot-specific concentration.
      Host Rabbit
Specificity Human Nestin. The antibody shows no reactivity to vimentin or GFAP.
      Isotype IgG
      Species Reactivity
      • Human
Species Reactivity Note Human. No reactivity to rat or mouse nestin. Other species have not yet been tested
      Antibody Type Polyclonal Antibody
      Entrez Gene Number
Entrez Gene Summary Nestin is an intermediate filament protein that was first identified with a monoclonal antibody by Hockfield and McKay (1985) [PubMed 4078630]. It is expressed predominantly in stem cells of the central nervous system in the neural tube. Upon terminal neural differentiation, nestin is downregulated and replaced by neurofilaments.[supplied by OMIM]
