Descripción Glial Derived Neurotrophic Factor Protein, Recombinant human
Visión de conjunto Human Glial Derived Neurotrophic Factor (GDNF) is a 30.4 kDa protein that specifically promotes dopamine uptake and survival of midbrain neurons. GDNF is a disulphide linked homodimeric protein consisting of two 134 amino acid chains.

Actividad específica Chemicon's human GDNF is fully biologically active when compared to standards as determined by the dose-dependent dopamine uptake by rat ventral mesencephalic cultures. The ED(50) was found to be in the range of 5-10 ng/mL. <br />The ED(50) as determined by the proliferation of rat C6 cells is ≤ 0.1 ng/mL, corresponding to aspecific activity of ≥ 1 x 107 units/mg.
      Número de Entrez Gene
Resumen de Entrez Gene This gene encodes a highly conserved neurotrophic factor. The recombinant form of this protein was shown to promote the survival and differentiation of dopaminergic neurons in culture, and was able to prevent apoptosis of motor neurons induced by axotomy. The encoded protein is processed to a mature secreted form that exists as a homodimer. The mature form of the protein is a ligand for the product of the RET (rearranged during transfection) protooncogene. In addition to the transcript encoding GDNF, two additional alternative transcripts encoding distinct proteins, referred to as astrocyte-derived trophic factors, have also been described. Mutations in this gene may be associated with Hirschsprung disease.

      Número UniProt
Resumen UniProt FUNCTION: SwissProt: P39905 # Neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.
SIZE: 211 amino acids; 23720 Da
      SUBUNIT: Homodimer; disulfide-linked.
      SUBCELLULAR LOCATION: Secreted.
TISSUE SPECIFICITY: In the brain, predominantly expressed in the striatum with highest levels in the caudate and lowest in the putamen.
DISEASE: SwissProt: P39905 # Defects in GDNF may be a cause of Hirschsprung disease (HSCR) [MIM:142623]. In association with mutations of RET gene, defects in GDNF may be involved in Hirschsprung disease. This genetic disorder of neural crest development is characterized by the absence of intramural ganglion cells in the hindgut, often resulting in intestinal obstruction. & Defects in GDNF are a cause of congenital central hypoventilation syndrome (CCHS) [MIM:209880]; also known as congenital failure of autonomic control or Ondine curse. CCHS is a rare disorder characterized by abnormal control of respiration in the absence of neuromuscular or lung disease, or an identifiable brain stem lesion. A deficiency in autonomic control of respiration results in inadequate or negligible ventilatory and arousal responses to hypercapnia and hypoxemia.
SIMILARITY: SwissProt: P39905 ## Belongs to the TGF-beta family. GDNF subfamily.

      Información de almacenamiento y transporte
Condiciones de almacenamiento The lyophilized powder, though stable at room temperature, is best stored desiccated below 0°C. The lyophilized power can be reconstituted to 0.1-1.0 mg/mL. This solution can be diluted into water or other buffered solutions. Reconstituted GDNF should be stored in undiluted aliquots at -20°C for up to 6 months.
