Two-parameter analysis can achieve target-specific detection of phosphorylation, and eliminates false positives in a mixed cell population. In an unstimulated system, only total H2A.X expression is seen (97.22%) in A. Upon treatment with 100 µM Etoposide for 2 hours, simultaneous measurement of both total and phospho H2A.X confirms target specificity (2.09% to 97.15%) B.
