High Cell Culture Yield

Yields of adherent cells grown in Millicell® HY cell culture flasks are linearly proportional to surface area and closely match theoretical yields.
CHO-k1 cells seeded at 40,000 cells/cm2, propagated with 0.2 mL/cm2 culture medium and incubated for 48 hours at 37 ºC, 6% CO2, 95% relative humidity. Cells were washed with 0.02% EDTA followed by harvest via enzyme dissociation for 10-15 minutes at 37 ºC. Cell counts were normalized to average cell yield from T75 flasks and reported as T75 flask equivalents.

Perfectly uniform cell growth on each layer for results you can count on:

Five times the surface area because of 5 identical layers.
Separating the layers of a 5-layer Millicell HY cell culture flask shows perfectly uniform cell growth across all 5 layers. Adherent cells were stained prior to flask disassembly.

Multiple Cell Types

HY flask demonstrates excellent recovery and viability across both adherent and suspension cells.

Cell yield and viability data comparing growth of six cell types in T75 flasks vs. Millicell HY cell culture flasks.
Cell were seeded at 20,000 – 40,000 cells/cm2 with 0.2 mL/cm2 media and incubated for 48 h at 37 ºC, 6% CO2, 95% relative humidity. Cells were washed with either PBS or 0.02% EDTA followed by harvest via enzyme dissociation for 10-15 min at 37 ºC. Cell count and viability measurements were performed using a guava® easyCyte™ flow cytometer with guava ViaCount® flex reagent. Theoretical cell yield = (cells recovered from one T75 flask) ÷ 75 cm2 x (total surface area in cm2).

Performance Comparison

HY flask produces better cell recovery compared to other multilayer cell culture flasks.
To compare cell yield between Millicell HY flasks and other multilayer cell culture flasks, CHO-k1 cells were seeded, recovered, and counted in experiments shown for High Yield. % cell yield = 100 x actual cell yield divided by theoretical cell yield (calculated for High Yield experiments).
